发酵茶中多种真菌毒素超高效液相色谱 ——串联质谱法的测定

      采用乙腈/水溶液(84+16)提取试样、MycoSpin 400多功能柱净化,建立了发酵茶(普洱茶、湖南黑茶和红茶)中黄曲霉毒素B1(AFB1)、黄曲霉毒素B2(AFB2)、黄曲霉毒素G1(AFG1)、黄曲霉毒素G2(AFG2)、赭曲霉毒素A (OTA)五种真菌毒素的简单、快速超高效液相色谱-串联质谱检测方法。试样以Agilent Zorbax Rrhd SB-C18柱分离,多反应监测(MRM)模式进行定量与定性分析,外标法定量。结果表明,在优化的条件下,五种真菌毒素在各自的线性响应范围内线性关系良好,相关系数(r)不低于0.9983,定量限(LOQ)为0.1~0.5 μg/kg,低、中、高3个加标水平的平均回收率(n=3)为63.5%~125.0%,相对标准偏差(RSD)为3.9%~17.2%。该方法前处理简单、检测速度快、净化效果好、结果准确、可靠,适合发酵茶中多种真菌毒素的快速检测。

      A method for the determination of five mycotoxins, AFB1、AFB2、AFG1、AFG2、OTA in fermented tea (Pu'er tea, and Hunan dark tea and black tea) was eatablished, by ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) under multi-reactions monitoring (MRM) models. The tea samples were extracted with 84% acetonitrile, then cleaned up with MycoSpin 400 multifunctional clean-up column. The mycotoxins were separated on a Agilent Zorbax Rrhd SB-C18 column ( 2.1 mm×100 mm,1.8μm) using (0.01% formic acid-0.05% ammonia-H2O)/methanol as mobile phase and detected by MS/MS. The mass spectrometry was conducted by positive (ESI+) and negative (ESI-) electrospray ionization with external standard method. The results indicated that the contents of five mycotoxins had good linear relationship with their peak areas, the correlation coefficients (r) were more than 0.9983, the limits of quantification (LOD)ranged from 0.1to 0.5μg/kg, and the spiked recoveries of five mycotoxins ranged from 63.5% to125.0%, at the spiked levels of 0.5~20μg/kg, the relative standard deviation (RSD, n=3) were between 3.9% and 17.2%. This method was rapid, simple, sensitive and reproducible, which could be used for the determination of multi-component mycotoxin contaminants in fermented tea.
